Synthesis and Solubility Evaluation of di-Mannich 2,6-Dimethylmorpholine Derivative of Curcumin Pyrazole

Abstract


Curcumin has various biological activity and relatively safe. However, it has poor water solubility, as a key factor of poor absorption following oral administration. In order to obtain a new curcumin derivative having better water solubility, di-Mannich derivative of curcumin pyrazole was synthesized by Mannich reaction of curcumin pyrazole, formaldehyde and 2,6-dimethylmorpholine, and the water solubility was evaluated. The structure of the synthesized compound was confirmed on the basis of IR, 1H-NMR, and 13C-NMR spectral data. The solubility evaluation revealed that the solubility of di-Mannich 2,6-dimethylmrpholine curcumin pyrazole in water (pH 7.4) was about three times  better than curcumin and curcumin pyrazole.
