Optimization of Microwave-Assisted Extraction From Suruhan (Peperomia pellucida (L.) Kunth) Herbs Of The Total Flavonoid Content and Antioxidant Activity

Abstract


Peperomia pellucida (L.) Kunth known as “ Suruhan” is one of the potential medical plants that used for the treatment of rheumatism, gout, headache and abdominal pain. Chemical constituens contained in this plant are alkaloids, flavonoid, sapponins tannins. The extraction method Microwave Assisted Extraction (MAE) which will be used to obtain the total flavonoid content, flavonoid profiles using thin layer chromatography (TLC) densitometry and test the antioxidant activity using DPPH optimum of Peperomia pellucida (L.) Kunth herb. This study to obtain optimum conditions MAE covering, solvent concentration, extraction time, solid-liquid ratio, and MAE power using RSM. The analysis result of optimum condition obtained was the content of flavonoid was 36,91 mg quercetin ekuivalents / g extract the condition with solvent concentration of ethanol 80 %, solid-liquid ratio 1:12, extraction time of 2 minutes, and MAE power 70%. The result of antioxidant activity 28.85 % with solvent concentration of ethanol 65%, solid-liquid ratio 1:10, extraction time 3 minutes and MAE power 50%. TLC densitometry profile showed flavonoid compounds contained in Peperomia pellucida (L.) Kunth herb. The analysis showed no correlation between the levels of total flavonoids with antioxidant activity.
