Anti-hepatocellular Carcinoma Activity Using Huh7it Cells of Several Plants From East Kalimantan, Indonesia

Abstract


Background: Hepatocellular carcinoma (HCC) was the sixth most common cancer worldwide with 746.000 cases of liver cancer related deaths in 2013. Hepatitis B or C virus infection and chronic alcohol consumption were some factor that increase the risk of developing HCC. Currently, the most common HCC treatment is a surgical therapy. However, success rates are low and often patients do not qualify. Therefore the investigation for alternatif treatment is greatly needed. Objective: This study was to determine anti-HCC activity of several plants belong to Rutaceae, Moraceae and Lauraceae family collected from Balikpapan, East Kalimantan. Materials and Methods: : Leaves and stem bark parts from eight different species of plants were extracted gradually using n-hexane, dichloromethane and methanol. Anti-HCC activity test was conducted using Huh7it cells by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay at single dose 30 µg/mL. Results: Four extracts out of 42 extracts showed more than 80% inhibition of HCC grow. The extracts were n-hexane (HLS-F) and dicloromethane extract of Luvunga scandens leaves (DLS-F), dicholoromethane extract of Artocarpus sericicarpus leaves (DAS-F), and dicloromethane extract of Artocarpus anisophyllus stem bark (DAA-SB) with % inhibition of 97.88 ± 0.80, 83.97 ± 1.02, 97.00 ± 0.44, and 87.64 ± 2.41, respectively. Conclusion: Based on the results of anti-HCC, this study demonstrates that 4 extracts i.e HLS-F, DLS-F, DAS-F, and DAA-SB had a higher anticancer activity compared with others extracts and possibility become good candidate to develop anti-HCC agents.
