Application of Natural Deep Eutectic Solvents (NADES) for Sappan Wood (Caesalpinia sappan L.) Extraction and Inhibition Test of DPP IV Activity

Abstract


ABSTRACT

The main compound in sappan wood (Caesalpinia sappan L.) namely brazilin has various pharmacological activities, one of them is hypoglycemic through inhibition of DPP IV activity. Natural Deep Eutectic Solvents (NADES) can be used as alternative solvents to replace organic solvents that are toxic and harmful to the environment. In this study NADES was used to extract brazilin from sappan wood and the extract was used to determine the inhibition of DPP IV activity. The selected NADES composition are choline chloride and betaine as hydrogen bond acceptors with glycerol and lactic acid as hydrogen bond donors. Extraction with NADES was carried out using Ultrasound Assisted Extraction (UAE) and brazilin level was measured using High Performance Liquid Chromatography (HPLC). Extraction was carried out under conditions of 50% water addition and 50 minutes extraction time. Choline chloride-glycerol is the best NADES for extracting brazilin with brazilin levels 114.04 mg/g compared to choline chloride-lactic acids (104.81 mg/g) and betaine-lactic acid (111.63 mg/g). Sappan wood extract (50 ppm concentration) which extracted using NADES choline chloride-glycerol has 84.24% inhibition value of DPP IV activity. Based on this study it can be concluded that choline chloride and betaine based NADES can extract brazilin from sappan wood and the extract can inhibit DPP IV activity.
