α-mangostin hydrogel film based chitosan-alginate for reccurent aphthous stomatitis

Abstract


Background: Many antiseptic, local anaesthetic, or corticosteroid drugs have been used for basic therapy of Recurrent Aphthous Stomatitis (RAS). However, each drugs have harmful side effects. α -Mangostin, a main compound of mangosteen (Garcinia mangostana L.) peel extract, has been known as effective wound healing agent. Hydrogel film applications can be used as dressings which designed to separate mucosal lesions from the oral environment, and significantly to improve the effectiveness of RAS therapy. Objective: The purpose of this study was to develop α-mangostin hydrogel film based chitosan-alginate (FH-Katin) for RAS. Materials and Methods: in silico study of FH-Katin was analysed by discovery studio visualizer and autodock. Preparation of FH Katin used the solvent evaporation method with variations of doses about 10 mg, 25 mg, 50 mg. The physicochemical characterization was observed by using Scanning Electron Microscopy (SEM), X-ray diffraction (XRD), differential scanning calorimetry (DSC). Furthermore, the effect of variations in a dose of α-mangostin on the characteristics of FH Katin was evaluated by swelling ratio, degradable, tensile strength and elongation at break followed by in vitro testing including the test of mucoadhesive time and release of α-mangostin. Results: The results of in silico study showed a hydrogen bonding between chitosan-alginate and α-mangostin. The results of SEM, DSC, and XRD indicated that the surface of FH-Katin was homogeneous and had a crystalline form of α-mangostin. FH-Katin with 50 mg of α mangostin significantly improved swelling ratio, tensile strength, and elongation at break compared to control. In addition, it was not degrade in 20% TCA solution and trypsin. The result of in vitro released study confirmed that the release of α mangostin from FH Katin was the Fickian diffusion model. Finally, the mucoadhesive study revealed that FH Katin had a good mucoadhesive properties Conclusion: These results suggest that hydrogel films based chitosan-alginate can be used as carriers of α mangostin for RAS therapy.
