Simultaneous Analytical Method of Retinoic Acid and Betamethasone Dipropionate in Cream Formula by High Performance Liquid Chromatography-Photodiode Array

Abstract


Retinoic acid (REA) could reduce the hyperkeratinization in the sebaceous follicle and accordingly decrease the sebum secretion and inflammation. Betamethasone dipropionate (BEP) is glucocorticosteroids with anti-inflammatory and immunosuppressive abilities, its applied as a topical cream such as eczema. This study aimed to develop simple, sensitive and validated a high performance liquid chromatography-photodiode array (HPLC-PDA) method for determination of REA and BEP simultaneously in cream as a topical formulation. The mixture of REA and BEP was separated on Bodapeck C18 5 μm column (4.6 mm × 150 mm), with PDA detector on 200-400 nm wavelength and flow rate at 1 mL/minutes. The mobile phase was methanol-buffer pH 5 (85:15). The retention time of REA and BEP was found to be 3.817 min and 1.902. The method was linear with r > 0.9990 over the range 0f 25-250 µg/ml for REA and BEP respectively. The recovery was acceptance criteria with the range of 99.05 – 100.96% for all component. The validated method was acceptance criteria, thus developed method can be used in routine analysis of REA and BEP in the cream formula.
