Neuroprotective Neolignan From the Bark of Magnolia Officinalis var. biloba

ABSTRACT
Background: The bark of Magnolia officinalis Rehd. et Wills var. biloba (M. officinalis var. biloba), important traditional Chinese medicines, is widely used in many prescriptions for the treatment of pain from abdominal distension, dyspepsia and phlegm. Objective: Phytochemistry on the ethanolic extract of the bark of M. officinalis var. biloba and Neuroprotection assay of New compounds from bark of M. officinalis var. biloba. Materials and Methods: The ethanolic extract of the bark of Magnolia officinalis var. biloba were subjected to various chromatographic techniques such as silica gel, sephadex LH-20, MPLC and preparative HPLC and the structures were elucidated on the basis of 1D and 2D NMR, HRESIMS and electronic circular dichroism (ECD), new compounds 1-2 were tested for the neuroprotective against oxygen glucose deprivation (OGD)-induced SK-N-SH cell injury in vitro. Results: New compounds 1-2 and 3-8 known compounds were isolated from the bark of M. officinalis var. biloba. At a concentration of 10 μM, compounds 1-2 increased OGD-induced SK-N-SH cell injury from 52.4% to 64.3%, 58.0%, respectively. Compound 1 more powerful than positive control drugs PHPB (59.5%). Conclusion: Based on these results, neuroprotective compounds 1-2 from the ethanolic extract of the bark of M. officinalis var. biloba make it can play potential roles in anti-dementia.
