Abstract
Objectives: Oxidative stress plays a major role in renal dysfunction caused by type 2 diabetes melitus. Up to now, there is no sensitive biomarker can be used in early detection of renal dysfunction. This study aims to analyze the difference and correlation between urinary hydrogen peroxide concentration as biomarker of oxidative stress with estimated Glomerular Filtration Rate as parameter of renal function in type 2 diabetic patients consuming biguanid and biguanid-sulfonylurea.
Methods : Urine and serum sample were collected from type 2 diabetic patients in Puskesmas Pasar Minggu. Concentrations of urinary hydrogen peroxide were measured using microplate reader with Ferrous Ion Oxidation Xylenol Orange 1 (FOX-1) method and normalize with urine creatinine that measured with kinetic Jaffe method. Value of eGFR were calculated using Cockroft-gault, MDRD, dan CKD-EPI formulation.
Results : Concentration of urinary hydrogen peroxide in two groups of treatment have no significant difference (p = 0,545), even with eGFR (Cockroft-Gault p = 0,832; MDRD p = 0,890; dan CKD-EPI p= 0,548). Results of correlation analysis between urinary hydrogen peroxide with eGFR also showed no significant correlation (Cockroft-Gault p = 0,916 ; MDRD p = 0,842; dan CKD-EPI p= 0,703). Biguanid and biguanid-sulfonylurea treatment didn’t show any significant difference in hydrogen peroxide level and eGFR in this study.
