Abstract
Transfersome has been widely used to increase the penetration of drugs derived from natural ingredients. One of the natural ingredient for health and cosmetics is green tea leaves extract (Camellia sinesis L Kuntze) containing catechin has potent antioxidant activity. Epigallocatechin gallate (EGCG) as one of the most dominant catechin compounds used as a marker analysis. However, EGCG has a large molecular weight and hydrophilic so that it’s difficult to penetrate through the skin. The aims of this study were to formulated and produced transfersome with good characteristics that increase penetrated through the skin. In this research, transfersome were formulated with different concentrations, equal to 1% (F1), 1.5% (F2), and 2% (F3) of EGCG and using thin layer hydration method. Transfersome then formulated into a gel and control gel prepared without transfersome. Both gels were evaluated and in vitro penetration tested using Franz diffusion cell with the skin of female Sprague Dawley rats. The result showed that F1 had the best formula with spherical shape, Dmean volume 107.82 ± 0.44, polidispersity index 0.07 ± 0.01, zeta potential -40.3 ± 0.10 mV, and entrapment efficiency 63.16±0.65%, Total cumulative amount of penetrated EGCG from transfersom gel was 1302.63 ± 20.67 μg/cm2 and the flux was 57.594 ± 0.91 μg.cm-2.hour-1. Total cumulative amount of penetrated EGCG from non-transfersom gel was 414,86 ± 4.40 μg/cm2 and the flux was 36.144 ± 1.22 μg.cm-2.hour-1. Based on these result it can be concluded that gel transfersome green tea leaves extract can increase penetration.
