Abstract
Analysis of Acrylamide in Dried Blood Spot of Students by Ultra High Performance Liquid Chromatography - Tandem Mass Spectrometry

Acrylamide is a chemical which is genotoxic, potentially carcinogenic in humans and is formed when starchy foods are fried or baked at temperature of more than 120oC. In this research, analysis of acrylamide was performed in dried blood spot (DBS) samples of 15 students, who often consume acrylamide-containing foods and 15 subjects of negative control group. The aim of this study is to know whether there is a significant difference between the concentration of acrylamide in the DBS samples of the students and the negative control subjects. DBS samples were extracted by a method of ultrasound-assisted liquid extraction and analyzed by using Ultra High Performance Liquid Chromatography – tandem Mass Spectrometry (UHPLC-MS/MS). The bioanalytical method of acrylamide in DBS samples of students with propranolol as the internal standard by using UHPLC-MS/MS had been partially validated in this study. The % diff value of the within-run accuracy obtained in this study were within ±20% for the LLOQ and within ±15% for the other QC samples. The coefficient of variation (CV) value of the within-run precision obtained were no more than 15% and no more than 20% for the LLOQ. Student who consumed acrylamide-containing foods the most frequently presented the highest level of acrylamide. The DBS samples of the negative control group presented the presence of acrylamide in very small concentration. The independent samples t test result showed that there was a significant difference in acrylamide concentration between the DBS samples of the students and the negative control subjects.
