Preparation of Gelatin-based Supramolecular Soft Capsule Films through Host-guest

Abstract


Background: Soft capsule is a film formed into a certain shape while filling it with a liquid or paste. It is widely used for medicines and supplements. On the other hand, to improve the quality of soft capsules prepared by a rotary method, enhancement of strength in the seam is required. To obtain enough strength in the seam of soft capsules, the development of novel materials that possess self-healing properties, elasticity, toughness, etc. are required. Meanwhile, various supramolecular polymeric materials have been developed through cyclodextrin (CyD)-based host-guest interactions, and these materials show above properties. Objective: In the present study, we synthesized b-CyD/gelatin conjugate (b-CyD-gelatin) and adamantane/gelatin conjugate (Ad-gelatin), and prepared a supramolecular gel by mixing the both components. In addition, we evaluated the potentials of the supramolecular gel as a soft capsule film. Materials and Methods: Ad-gelatin and b-CyD-gelatin were synthe-sized through the activation of carboxyl group of 1-adamantanacetic acid or glucuronylglucosyl-b-CyD (GUG-b-CyD) with N-hydroxysuccinimide and following reaction with the amino group of gelatin. Ad-gelatin/b-CyD-gelatin hydrogel was obtained by heating the solution of Ad-gelatin/b-CyD-gelatin at 80ºC for 4 h, and then cool-ing at 4ºC. Mechanical properties of Ad-gelatin/b-CyD-gelatin hydrogel were evaluated using a tensile tester. Results: According to the results of 1H-NMR spectrum, the preparation of b-CyD-gelatin and Ad-gelatin was confirmed. The solution of Ad-gelatin/b-CyD-gelatin showed higher viscosity than that of gelatin alone. Ad-gelatin/b-CyD-gelatin hydrogel showed higher extensibility and strength than gelatin alone. In addition, the hy-drogel possessed self-healing property. Conclusion: Ad-gelatin/b-CyD-gelatin hydrogel has the potentials as an advanced soft capsule film.
