Inhibition Test of Elastase Activity in Macassar Kernels (Rhus javanica L)

Abstract


This study aims to conduct the inhibition of elastase activity from several parts of the plant of Macassar Kernels (Rhus javanica L), namely the stem, leave, greenish fruit and blackish-grey fruit. The extraction was prepared by maceration with ethanol 96 % to obtain the ethanolic extract. The parts of the plant were analyzed for chemical compounds.Ethanolic leaves extract of Macassar Kernels (EL) contains flavonoids, tannins, terpenoids, and glycosides. Ethanolic stem extract of Macassar Kernels (ES) stem contains  flavonoids, tannins, and terpenoids.  Ethanolic greenish and blackish-grey fruit extract contains tannins, terpenoids, and steroid.The elastase inhibition activity test was carried out with a Versamax microplate reader as a device, N-Succ- (Ala) 3-nitroanilide (SANA) as a substrate, and elastase enzyme derived from porcine pancreas. The percentage of inhibition from 100 ppm ethanolic extract was obtained by 12.30% (EL), 45.30% (ES), and there was no inhibition in the fruit parts. Ethanolic stem extract of Macassar Kernels has the highest potential in elastase inhibitory activity.
