Pharmacognostical Evaluation of Kadamba (Mitragyna speciose [Koth.] Havil.) leaves from East Kalimantan Indonesia

Abstract


Background: Mitragyna speciosa (Korth.) Havil. (Rubiaceae family) is known locally “Kadamba”. It is planted grows in the wild of Kalimantan island, mainly in East Kalimantan. It is native to Southeast Asian, including Indonesia, Malaysia, the Philippines, Thailand, and Papua New Guinea. Objective: The present study aimed to evaluate the pharmacognostical characteristic of Kadamba (M. speciose) leave from East Kalimantan Indonesia. Method: using several characteristic tests involve macroscopic and microscopic analysis, fluorescence test, phytochemical and physicochemical properties. Result: The macroscopic of M speciosa fresh leaves are shiny and dark green, ovoid-accumulated which have an average size of more than 14-20 cm (5.5-7.7 in) long and 7-12 cm wide (2.8-4.7 in) with 12-17 vein pairs, and opposite in growth patterns. The microscopic characteristic showed epidermis cells, parasitic stomata, glandular trichomes, and vascular bundle. Fluorescence test shows the characteristics of secondary metabolites contained in the sample based on their ability to fluorine on UV rays after the addition of specific reagents. Phytochemical showed the presence of some secondary metabolites (mainly alkaloid, flavonoid, polyphenols). The physicochemical characters showed a water content of 10.4%.
