DETERMINATION OF FLAVONOID TOTALS OF ETHANOL EXTRACTS, ETHYL ACETATE FRACTIONS AND n-BUTHANOL FRACTION OF PASAK BUMI (Eurycoma longifolia Jack.) BORNEO USING FTIR SPECTROSCOPY AND PLSR

Abstract


FTIR and chemometrics methods have been used to analyze flavonoid levels from various plants, one of them is pasak bumi (Eurycoma longifolia Jack.). Result of data analysis can be accepted and accounted for if they have been validated. This study is aimed to determine the level of flavonoids and validate the analytical method of flavonoids levels of ethanol extract, ethyl acetate fraction and n-butanol fraction of pasak bumi root (Eurycoma longifolia Jack.) using spectroscopy FTIR and Partial Least Square Regression (PLSR) . The analysis was carried out by PLSR to make a prediction model of the levels of flavonoids in ethanol extract, ethyl acetate fraction and n-buthanol fraction of pasak bumi roots. The result of prediction model had the equation of y = 0.9996x + 0.0004 and tested on the sample and obtained the total flavonoid content from the predictions of E. longifolia root extract from Mandiangin, Condong and Sabuai at 0.4915; 04998 and 0.5324 ( b/b). Flavonoid content of the total prediction of ethyl acetate fractions of E. longifolia root Mandiangin, Condong and Sabuai was 1.9941; 2.2587 and 2.3213 (%b/b). Flavonoid content in total prediction of n-buthanol fraction of E. longifolia root Mandiangin, Condong and Sabuai was 0.7876; 1.1144 and 0.5423 (%b/b). The results of the validation test obtained LOD and LOQ values at 0.0417 (%b/b) and 0.1390 (%b/b). The result of linearity test was R2 = 0.9998. The value of precision and accuracy expressed in % RSD and % recovery met the requirements, which was ≤ 2% and 98-102%.
