Comparison of Solvent Concentration for Extraction of Sulfhydryl-Containing Compound in Leucaena leucocephala (Lam.) de Wit and Other Leguminos Seeds

Abstract


Background: Leucaena leucocephala (Lam.) de Wit, Parkia speciose Hassk and Archidendron jirringa (Jack) I.C. Nielsen seeds extract have therapeutic properties from sulfhydryl-containing compounds. Objective: The main objective of this research was to compare the effect of solvent concentration for extraction of leguminos seeds in term of yield and sulfhydryl compound levels. Materials and Methods: Seeds macerated in room temperature (250C) and ratio solid and solvent (1:10). The solvent concentration used was 30%, 50%, 70% and 96% ethanol. The yield of extract stated in ratio weight of extract to simplicias.  Results: Sulfhydryl compound levels was determined by Ellman reagent and the reaction product was analysed using spectrophotometer at 535 nm wavelength. The 96% ethanol solvent produced the highest yield extract and sulfhydryl compound levels was significantly different with the other concentrations of ethanol.  Archidendron jirringa (Jack) I.C. Nielsen seeds have the highest concentration of sulfhydryl compound compared with other leguminos seeds. Conclusion: The result showed that the solvent concentration had effect for extraction of sulfhydryl-containing compound in leguminos seeds.
